Charlene Lucille de Carvalho-Heineken was born on June 30, 1954, in the Netherlands. Her father was Freddy Heineken, a prominent Dutch industrialist and the owner of Heineken N.V. Her mother, Lucille Cummins, was an American from a family involved in bourbon whiskey distilling. She was educated at Rijnlands Lyceum Wassenaar and later studied law at Leiden University.

Charlene de Carvalho-Heineken inherited a 25% controlling stake in Heineken N.V. in 2002 after her father's death, propelling her onto the world stage. She is also an executive director of the company.
